I am already working on a Post-Graduation Work Permit in Canada. Do I still need a Labour Market Impact Assessment?

It depends on your specific situation and the immigration program you are invited to apply for under Canada Express Entry. If you are in Canada on a Post-Graduation Work Permit and have a valid job offer, the employer may still need to get an LMIA to ensure you qualify for the Federal Skilled Worker or Federal Skilled Trades Program. The only time the LMIA is not needed is when the position being offered is exempt. If you are invited to apply through the Canadian Experience Class, no LMIA is needed. Under Canada Express Entry, there are 200 or 50 points available for a valid job offer supported by an LMIA.
